Complete nucleotide sequence and ambisense genomic organization of M RNA of watermelon silver mottle tospovirus (WSMV)

Abstract: To verify the taxonomic position of WSMV, a new member of the genus Tospovirus in the family Bunyaviridae, the nucleotide sequence of its M RNA was determined and compared with that of other tospoviruses. The complete nucleotide sequence of WSMV M RNA was 4880 nt in length with two open-reading frames (ORFs) in an ambisense organization. The NSm ORF located on the viral strand of the M RNA contained 936 nt, coding for a protein of 312 amino acids. Comparison of the WSMV NSm protein with those of impatiens necrotic spot virus (INSV) and tomato spotted wilt virus (TSWV) revealed that the amino acid similarities were 51.0% and 43.3%, respectively. The G1/G2 ORF located on the viral complementary strand of the M RNA contained 3366 nt, coding for a protein of 1121 amino acids. Comparison of the WSMV G1/G2 precursor protein with those of INSV and TSWV revealed that the amino acid similarities were 42.4% and 46.5%, respectively. The two ORFs were separated by a 473 nt A-U-rich intergenic region with nucleotide identities of 59.0% and 50.3% to those of INSV and TSWV, respectively. The 5' and 3' non-coding regions of WSMV M RNA contained 55 and 47 nt, respectively, were shorter than those of INSV and TSWV, and shared 78.2-85.5% and 76.6-78.7% identity, respectively. Comparison of M RNAs of WSMV, INSV, and TSWV revealed a consensus sequence of 8 nt of 5-AGAGCAAU....-3' at their 5' ends and 5'-.....AUUGCUCU-3' at their 3' ends. These results strengthened the belief that WSMV belongs to the Tospovirus genus, as a new member phylogenetically distinct from INSV and TSWV.
