Proteome analysis of the interaction among Trichoderma, lettuce and pathogenic fungi

Abstract: Fungi of the genus Trichoderma are among the most effective bio-pesticides used for controlling diseases caused by pathogenic fungi to vegetable crops. Molecular activation of specific genes in the antagonist occurs during its interaction with pathogen and plants. The spectrum of enzymes involved in biological control activity includes glucanases, chitinases, lipases and proteases. In order to identify and characterize new biocontrol proteins, we produced proteomic maps of the in vivo interaction among T. harzianum (strain T22), Lactuca sativa and the pathogens Rhizoctonia solani and Sclerotinia sclerotiorum. To extract intracellular proteins (ICs) from the antagonist's mycelium during the threecomponent interaction, T. harzianum was grown in the presence of lettuce seedlings immersed in water agar and by separating the plants from the mycelium with a cellophane membrane; the fungal pathogens were also included in the system. Extracellular proteins (ECs) were extracted from culture filtrates obtained by coculturing plant, pathogen and antagonist in a two phase (solid-liquid) system. ICs and ECs proteins were analysed by two-dimensional electrophoresis (2-DE) and qualitative and quantitative differences were detected. Spots corresponding to proteins differentially expressed were identified by matrix-assisted laser desorption ionisation-time of flight (MALDI-TOF) mass spectrometry (MS).
