Characterization of Colletotrichum species associated with coffee berries in northern Thailand

Abstract: The identity of Colletotrichum species on Coffea (coffee) is not clearly understood. We report on Colletotrichum species associated with coffee berries in northern Thailand and compare them to species reported to cause coffee berry disease elsewhere. Morphological, cultural, biochemical and pathogenic characters, together with DNA sequence analyses resulted in the isolates clustering into three species: Colletotrichum asianum sp. nov., C. fructicola sp. nov. and C. siamense sp. nov. Phylogeny inferred from combined datasets of partial actin, β-tubulin (tub2), calmodulin, glutamine synthetase, glyceraldehyde-3-phosphate dehydrogenase genes and the complete rDNA ITS1-5.8S-ITS2 regions revealed groupings that are congruent with morphological characters. Biochemical and DNA sequence data from multi-genes used here provided reliable data to differentiate between C. kahawae, C. gloeosporioides and the new Colletotrichum species.
