Abstract: Distribution of wheat streak mosaic virus (WSMV) in winter wheat plants was investigated in four winter wheat genotypes which were mechanically inoculated at the two leaf stage. Virus titer was measured with PA-ELISA. Distribution of WSMV was similar for all genotypes, i.e. virus titers in leaves were always higher than in sheaths and higher in sheaths than in roots. For Arapahoe and WGRC 27, virus titers in leaves and sheaths at 2 weeks post-inoculation (WPI) were higher than that at 4 WPI, while virus titers in roots at 2 WPI were lower than that at 4 WPI. Virus titer in Arlin leaves, sheaths and roots at 2 WPI was lower than at 4 WPI. For Vista, virus titers did not differ greatly between 2 and 4 WPI. At 2 WPI, the highest WSMV titer occurred in the lowest leaf for Arlin and Vista and in the second lowest leaf for Arapahoe and WGRC 27. At 4 WPI, the highest WSMV titer occurred in the second lowest leaf. At both 2 and 4 WPIs, the lowest virus titer occurred in the youngest leaf for all genotypes.
