Using RAPD analsis to assess the genetic diversity among races of Ustilago hordei and the heredity of genetic markers

Abstract: Pathogenicity on eight barley differential cultivars and random amplified polymorphic DNA (RAPD) analysis were used to evaluate genetic diversity among 14 races of Ustilago hordei. One hundred and twenty oligonucleotide primers were tested with basidiospore isolates from four races of U. hordei. Oligonucleotide primers that expressed polymorphism were selected and used to characterize a total of 14 races of U. hordei. Comparisons also were made among different isolates within each race. Polymorphism was observed among the races but not within each race. Two crosses were made between race 8 and race 14. Progeny were evaluated using the same selected primers to determine the heredity of these genetic markers.
