Variation of RAPD profiles in different geographical Indian populations of Heterodera cajani (Nematoda)

Abstract: The pigeonpea cyst nematode, Heterodera cajani, is an important pathogen of pulse crops in India, causing significant economic losses. Variations have been noticed in host preferences of different populations of the nematode. Genetic variation among eleven populations of H. cajani in India from major pigeonpea growing states was demonstrated by random amplified polymorphic DNA (RAPD)-polymerase chain analysis (PCR). 13 Operon random primers used in RAPD-PCR gave 296 scoreable markers with fragment size ranging from 200 to 3500 base pairs. The value of Jaccard's similarity coefficient ranged from 0.33 to 0.75. Overall, two distinct clusters were identified at an average similarity coefficient of 0.4. Cluster 1 comprising of two subclusters and 3 out groups included populations belonging to Hyderabad, Coimbatore, Allahabad, Bahadurgarh. Kanpur, Meja and Ghaziabad. Cluster 2 consisting of one subcluster and two out groups comprised of 4 populations belonging to Gulberga, Hisar, Kanpur (a different population) and Delhi. Principal component analysis also exhibited similar clustering pattern,. The bootstrap analysis indicated high confidence limits with values ranging from 50 - 100 in the branches generated by UPGMA clustering. There was no relationship between the observed genetic diversity and the geographical origin of the populations. The present findings may assist in diagnostics and breeding resistant pulse varieties.
