Host nuclear migration during incompatible and compatible interactions with Collectotrichum species

Abstract: Fluorescence microscopy was used to observe nuclear migration in living epidermal cells of Nicotiana tabacum L. cv. Xanthi following inoculation with Colletotrichum destructivum O'Gara, which is compatible with tobacco, and C. graminicola (Ces.) G.W. Wils., which is incompatible. The host had been transformed with a gene expressing a chimeric protein with a nuclear localization signal and green fluorescent protein (GFP), resulting in GFP marked nuclei. The epidermal cell nucleus migrated directly below the appressorium of C. graminicola, followed by movement back to the center of the cell after 40 h as the fungus failed to penetrate. The nucleus also migrated to just underneath the appressorium of C. destructivum, but stayed there during penetration. The nucleus remained adjacent to the tip of the multi-lobed vesicle as it grew biotrophically in the epidermal cell. Near the end of this period, approximately 10% of host nuclei moved away from the vesicles. Necrotrophic thin hyphae subsequently grew from the tips of the vesicles, host cellular disruption occurred, and host nuclei were no longer observed. Some nuclei appeared to initially contract from a rounded to a half-rounded or oval shape. Host nuclear movements suggest signaling between the fungus and its host that may be important in enabling a fungus to feed biotrophically without eliciting effective host defenses.
